Fluorescence

Fluorescence Spectroscopy is an optical technique that measures the emission of light from a sample following excitation by a specific wavelength. It provides highly sensitive, non-destructive analysis of molecular structure, concentration and dynamics. Applications include protein characterization, DNA analysis, environmental monitoring, pharmaceutical quality control and cellular imaging. Fluorescence instruments range from compact spectrofluorometers to advanced time-resolved and multiphoton systems.

How it works

FROM PRINCIPLE TO APPLICATION

A broadband light source (typically a xenon arc lamp) is spectrally filtered by a scanning excitation monochromator and focused onto the sample. Emitted fluorescence is collected at 90° to the excitation beam and dispersed by an emission monochromator onto a sensitive detector. By scanning both excitation and emission wavelengths, a complete excitation-emission matrix (EEM) is acquired. Time-resolved detection using time-correlated single photon counting (TCSPC) measures fluorescence decay lifetimes from picoseconds to microseconds, revealing molecular dynamics and environment.
