The HORIBA Duetta is a dual-function spectrometer combining UV-Vis absorbance and fluorescence emission measurement in a single compact benchtop instrument, designed for researchers who need both measurement modalities without purchasing and operating two separate instruments. The defining feature of the Duetta is its ability to simultaneously acquire absorbance and fluorescence emission spectra in a single measurement, which enables automatic inner filter effect (IFE) correction to be applied to fluorescence emission data. The inner filter effect is a common source of error in fluorescence measurements of concentrated or highly absorbing samples – when both the excitation and emission beams are attenuated by the sample, the measured fluorescence intensity is lower than the true value. By measuring absorbance and fluorescence simultaneously from the same sample aliquot, the Duetta can apply mathematically rigorous IFE correction in real time, giving accurate and quantitative fluorescence data even for concentrated samples without dilution. The instrument uses a CCD array detector for simultaneous fluorescence emission spectral detection, giving fast spectral acquisition across the full wavelength range. The Duetta covers a spectral range from approximately 200-1100 nm for absorbance and 350-1100 nm for fluorescence, making it suitable for visible and NIR fluorescent species including quantum dots, organic dyes, and fluorescent proteins.
