The Bio-Logic MOS-DAD is a diode array detector module for stopped-flow kinetics that records complete UV/Vis spectra (200-800 nm) at every time point, enabling simultaneous multi-wavelength kinetics and spectral evolution analysis of complex biochemical reactions.
The MOS-DAD (Multi-wavelength Optical System – Diode Array Detector) from Bio-Logic is the spectral detection module for the SFM stopped-flow platform, recording complete absorption spectra at each time point rather than single-wavelength traces, enabling 3D kinetic spectral analysis.
Spectral acquisition:
– Wavelength range: 200-800 nm (full UV/Vis)
– Detector: 256-element or 1024-element silicon diode array
– Spectral resolution: ~1 nm per pixel
– Acquisition rate: up to 1000 spectra/second
– Each spectrum: complete 200-800 nm scan in <1 ms
Advantages over single-wavelength detection:
- No need to select monitoring wavelength in advance
- Detect multiple intermediates simultaneously at their optimal wavelengths
- Global spectral analysis: decompose into pure intermediate spectra
- Identify unexpected intermediates not visible at a single wavelength
- Validate stoichiometry of chromophore changes
Data analysis:
- 3D dataset: time x wavelength x absorbance
- Global analysis software: SVD decomposition, target analysis
- Extract species-associated difference spectra (SADS)
- Kinetic traces extractable at any wavelength post-acquisition
- BioKine32 integration for acquisition; compatible with third-party global analysis
Applications:
- Heme protein reactions: track Soret and Q-band changes simultaneously
- Flavin/cofactor enzymes: monitor all chromophores at once
- Protein conformational changes: UV/Vis spectral shifts
- Photochemical reactions: track all photoproducts
- Multicomponent reaction systems: resolve overlapping spectra
