The HORIBA OpenPlex is a surface plasmon resonance imaging (SPRi) system designed for real-time, label-free detection of biomolecular interactions in a massively parallel format. Unlike conventional single-channel SPR instruments that measure one interaction at a time, the OpenPlex uses a wide-field optical imaging approach to monitor hundreds of interaction spots simultaneously on a single gold-coated biochip. Surface plasmon resonance is a phenomenon where light excites surface plasmons at a metal-dielectric interface at a specific angle (the SPR angle), which shifts when molecules bind to the surface. By imaging the entire chip surface with a CCD camera, the OpenPlex captures binding kinetics across the entire array in real time – producing association and dissociation curves for every spot simultaneously. The OpenPlex biochips can be functionalized with different ligands at different spots using the SPRi-Arrayer, enabling multiplexed screening of many analyte-ligand pairs in a single injection cycle. This multiplexed capability is particularly valuable for drug discovery screening (fragment-based screening, hit-to-lead validation), biomarker discovery in serum or complex biological matrices, antibody characterization (specificity, cross-reactivity), and detection of pathogens or environmental contaminants. The instrument interfaces with microfluidic sample injection for controlled analyte delivery and is compatible with standard SPRi protocols. HORIBA’s SPRi-Lab+ software provides real-time imaging, kinetics extraction (ka, kd, KD), and multi-curve analysis tools.
